I've never taken the Chinatown bus. We're up by the GW Bridge, so getting down to Chinatown on the subway would take at least an hour before we even got on the bus and took off.
I did recently take Greyhound (conveniently leaving from the Port Authority at 42nd Street) to Boston, and it was okay. It was really inexpensive--I think it was $20 one way, and then for an extra $5, you get advance seating so that you can be sure your whole party sits together. This was WELL worth it. The bus was clean and comfortable.
Then we blew out all the stops by taking the LimoLiner home--that was really nice!

There are a couple of new bus companies that started the Boston/NYC route in the spring. I kept the newspaper article, but I haven't checked to see if the companies are still in business. We're thinking of taking the bus for our visit to Manhattan next year.
