If you are flying to Key West, just head directly there. She'll think the trip is to Key West and be surprised when you go to the island.
If you are planning to fly to Miami and drive to Key West, go Friday, stay one night in Miami, then drive to Key West, it'll be a three stage surprise.
If you do decide to drive to Key West, I recommend only driving one way and flying back directly from Key West. The drive is long, and won't be very interesting on the way back, expecially when you'll have to give up time on the island to take it, and will be driving all that way just to get on a plane.
Obviously, if it were up to me, I'd fly to Key West. The plane from Miami is small (at largest only about 20 passengers) and the flight is short and kind of fun (if you like small planes). On the way back last time, we were in a plane so small it was just four passengers and the pilot, like a private jet.
